Item Open Access (1)H NMR-based metabolomics of Daphnia magna responses after sub-lethal exposure to triclosan, carbamazepine and ibuprofen(Elsevier Inc., 2016-09) Kovacevic, Vera; Simpson, André J.; Simpson, Myrna J.Pharmaceuticals and personal care products are a class of emerging contaminants that are present in wastewater effluents, surface water, and groundwater around the world. There is a need to determine rapid and reliable bioindicators of exposure and the toxic mode of action of these contaminants to aquatic organisms. (1)H nuclear magnetic resonance (NMR)-based metabolomics in combination with multivariate statistical analysis was used to determine the metabolic profile of Daphnia magna after exposure to a range of sub-lethal concentrations of triclosan (6.25-100μg/L), carbamazepine (1.75-14mg/L) and ibuprofen (1.75-14mg/L) for 48h. Sub-lethal triclosan exposure suggested a general oxidative stress condition and the branched-chain amino acids, glutamine, glutamate, and methionine emerged as potential bioindicators. The aromatic amino acids, serine, glycine and alanine are potential bioindicators for sub-lethal carbamazepine exposure that may have altered energy metabolism. The potential bioindicators for sub-lethal ibuprofen exposure are serine, methionine, lysine, arginine and leucine, which showed a concentration-dependent response. The differences in the metabolic changes were related to the dissimilar modes of toxicity of triclosan, carbamazepine and ibuprofen. Investigators suggested that changes in d15N values measured in urine, hair, and blood may be a good indicator of lean-tissue losses. During our investigations into the effects of population density on body condition and reproduction of female Arctic ground squirrels (Spermophilus parryii plesius) near Kluane Lake, Yukon, Canada, we examined the relations between body condition and d15N values. Data obtained from 20 livetrapped female ground squirrels suggested that reproductive females from a population with moderate density and low food availability experienced a reduction in body condition, as indicated by mass loss and changes in blood urea nitrogen (BUN) and glucose concentrations. In contrast, those from a population that failed to reproduce successfully and had high density and low food availability experienced no nutritional stress. Similarly, those females from a high-density population with high food availability (i.e., supplemented food) that reproduced successfully suffered no noticeable nutritional stress. In contrast to our prediction, d15N values did not show a decline with increasing body mass, and animals in poor and excellent body condition had similar d15N values. In addition, female ground squirrels from the same group with access to similar types of food (natural or supplemented) and with similar body masses, BUN, and blood glucose concentrations showed a difference of up to 1.8‰ in d15N values. Thus, our results suggest that the ecological process (i.e., diet selection) may have obscured the physiological one (i.e., recycling of nitrogen). Twenty AD patients were studied after obtaining both their informed consent and their caregivers'. They were divided into two groups of ten patients each one: Rivastigmine Group (RG) and Control Group (CG), and matched by age and sex. RG took Rivastigmine during 16 weeks (1.5 mg/day the first two weeks, 3 mg/day the following six weeks, and 4.5 mg/day the last eight weeks). CG took placebo during 16 weeks. Mini-Mental State Examination (MMSE), Magnetic Resonance Imaging (MRI), and MRS were performed before and after the 16-week study. MRI and MRS were carried out on a 1.5 Tesla system (Sonata, Siemens Erlangen®). Single Voxel Spectroscopy (1.8 cm x 1.8 cm x 1.8 cm = 5.83 cm3) were acquired with a 90°-180°-180° Spin-Echo Sequence (TR=1500 msec, TE=30 msec,192 averaged acquisitions). N-Acetylaspartate (NAA), Creatine (Cr), Choline (Cho), and myo-inositol (mI) intensity signals were detected and NAA/Cr, mI/Cr, Cho/Cr and NAA/mI ratios were calculated. Analysis of Variance was applied to the results. Tukey test was used for comparisons among ratio means with Bonferroni correction for small samples. Spearman rank correlation coefficients were calculated for both MMSE scores and measured metabolite ratios. A significant increase (p<0.05) in the NAA/Cr ratio for the Right FL and a significant increase (p<0.05) in the NAA/mI ratio for the Left TL were found in the RG after 16 weeks of Rivastigmine treatment. These changes were correlated to the RG patients' improvement in cognitive mental status that was uncovered through the MMSE's AD Assessment Scale Scores. No significant changes were found in the CG after 16 weeks of placebo treatment. MRS may provide a useful tool for monitoring the therapeutic response of AD patients to Rivastigmine. The majority of strains did not show high levels of homology to the first three genes of the 2,4-D degradation pathway, tfdA, -B, and -C. Most strains showed combinations of tfdA-, B-, and C- like elements that exhibited various degrees of homology to the gene probes. Strains having the same genomic fingerprints (as determined by repetitive extragenic palindromic PCR) exhibited the same hybridization pattern regardless of the geographic origin of the strain, with the exception of a strain isolated from Puerto Rico. This strain had the same genomic fingerprint as that of numerous other strains in the collection but differed in its hybridization against the tfdA gene probe. Members of the beta subdivision of the Proteobacteria class, specifically Alcaligenes, Burkholderia, and Rhodoferax species, carried DNA fragments with 60% or more sequence similarity to tfdA of pJP4, and most carried fragments showing at least 60% homology to tfdB. However, many strains did not hybridize with tfdC, although they exhibited chlorocatechol dioxygenase activity. Members of the alpha subdivision of the Proteobacteria class, mostly of the genus Sphingomonas, did not hybridize to either tfdA or tfdC, but some hybridized at low stringency to tfdB. The data suggest that extensive interspecies transfer of a variety of homologous degradative genes has been involved in the evolution of 2,4-D-degrading bacteria.Item Open Access 2.
